None of these five fell out of the sky. These are the labs that found them, the clinics that tested them, and the regulators that set the limits on how far the claims could go.
Former Pfizer Discovery Laboratories

The Kent site where sildenafil started out as UK-92,480, a hopeful angina candidate. The heart numbers were dull; the notes patients kept making about erections were not, and Pfizer turned the whole programme toward erectile dysfunction. Most of the modern PDE5 story opens on this road.

The Washington biotech that worked out tadalafil and partnered with Eli Lilly to carry it through late trials. Its molecule left the body far more slowly than its rivals, and that one pharmacokinetic quirk became Cialis — the long-window tablet the category now measures itself against.

The Wuppertal laboratories behind vardenafil, later sold as Levitra and the orodispersible Staxyn. Its compound is the most potent PDE5 inhibitor per milligram of the three, which is why the labelled doses read so much smaller than sildenafil's.

The New Jersey labs that developed finasteride, first as the prostate drug Proscar and then, at a fifth of the dose, as Propecia for male-pattern hair loss. The insight came from studying men born without working 5-alpha-reductase — a natural experiment that pointed straight at the enzyme.
Beecham Research Laboratories (historic)

The Surrey research site where Beecham chemists built the semisynthetic penicillins, amoxicillin among them, in the early 1970s. Adding a single hydroxyl group to ampicillin gave a molecule the gut absorbs far better — the reason amoxicillin, not ampicillin, sits in most medicine cabinets today.

Why the history matters
Provenance is not trivia. Sildenafil's start as a failed angina candidate explains both its cardiovascular cautions and its accidental fame; tadalafil's slow clearance out of the ICOS labs is the reason a "weekend" tablet exists; vardenafil's potency out of Wuppertal is why its labelled doses look so small. Finasteride came from studying men born without a working enzyme, which is why it targets one so precisely. And amoxicillin exists because Beecham chemists added a single hydroxyl group to ampicillin and made a molecule the gut actually absorbs.
